2011, Brain ResearchCitation Excerpt :To examine the effect that TNF-α had on CP-AMPARs in cerebellar Purkinje neurons, we took advantage of the selective permeability of cobalt ions. It is well accepted that cobalt influx selectively occurs through only GluR-2 lacking (calcium permeable) AMPA receptors and not NMDA or voltage dependent calcium channels (Pruss et al., 1991; Albuquerque et al., 1999; Sanchez et al., 2001; Deng et al., 2006). Through a modified cobalt uptake protocol, coupled with Timm's staining, we demonstrated that TNF-α drastically increases the total apparent calcium influx into Purkinje neurons through CP-AMPARs in response to activation by AMPA.
