Somatotopic reciprocal connections between the somatosensory cortex and the thalamic Po nucleus in the rat

Abstract

Retrogradely labeled neurons are observed in the posterior group of the thalamus (Po) after injection of wheatgerm agglutinin-horseradish peroxidase in the rat somatosensory cortex. These neurons are organized in rods elongated rostrocaudally, defining a clear somatotopic map. Injections of tritiated leucine in the somatosensory cortex indicate that these somatotopically organized connections are reciprocal. Injections of tritiated leucine in the dorsal column nuclei label afferent fibers in a small area dorsal to Po but not in the core of the nucleus. Po does not receive direct projections of ascending somatosensory afferents. It is hypothesized that this thalamic area participates in a thalamo-cortico-thalamic loop.

    Compared with the previous studies, one of the strengths of the present study is that the descending pathways from the S1 and S2 to the thalamus were also taken into account in the constructed DCMs, enabling the investigation of possible corticothalamic feedback regulations within this ‘thalamus-S1-S2’ network. Indeed, the existence of corticothalamic pathways have been reported in abundant anatomical studies in animals showing that both S1 and S2 send fibers to various thalamic nuclei especially the posterior nuclei of the thalamus (Auer, 1956; Chmielowska et al., 1989; Jones and Powell, 1968, 1970; Liao et al., 2010; Niimi et al., 1963; Nothias et al., 1988; Rinvik, 1968a, b; Spreafico et al., 1987; Stratford, 1954; Veinante et al., 2000). Anatomical evidence showed that corticothalamic projections are ~10 times more numerous than thalamocortical projections (Liu et al., 1995).

    In general, the whole VB has a somatotopic arrangement (Emmers, 1965; Saporta and Kruger, 1977; Waite, 1973), with a primary somatotopic map containing unilateral representations and a secondary with bilateral projections (Emmers, 1965). POm has also been shown to be somatotopic (Fabri and Burton, 1991; Nothias et al., 1988; Ohno et al., 2012), with cortical driver projections from L5b innervating four distinctly oriented somatotopic maps in different subdivisions of this nucleus (Sumser et al., 2017). Early investigations have shown a clearer arrangement in VB than in Rt and POm (Hoogland et al., 1987).

    Further, the reciprocal connections between the S1 and the ventroposterior thalamic nuclei have been well documented (Diamond et al., 1969; Jones et al., 1979; Mayner and Kaas, 1986; Hoogland et al., 1987; Land et al., 1995; Deschênes et al., 1998). In the present study, BDA-labeled axon terminals from the five orofacial S1 areas were somatotopically distributed in the ventroposterior medial thalamic nucleus (data not shown), whose somatotopy seemed to correspond to that proposed in earlier studies (Saporta and Kruger, 1977; Nothias et al., 1988; Fabri and Burton, 1991b; Liao et al., 2010). Therefore, it is highly likely that the orofacial S1 areas produce feedback regulation of the ascending somatosensory information to themselves in the third order somatosensory neurons in the ventroposterior medial thalamic nucleus as well as in the second-order ones.
