A new and rapid colorimetric determination of acetylcholinesterase activity
Abstract
A photometric method for determining acetylcholinesterase activity of tissue extracts, homogenates, cell suspensions, etc., has been described. The enzyme activity is measured by following the increase of yellow color produced from thiocholine when it reacts with dithiobisnitrobenzoate ion. It is based on coupling of these reactions: The latter reaction is rapid and the assay is sensitive (i.e. a 10 μ1 sample of blood is adequate). The use of a recorder has been most helpful, but is not essential. The method has been used to study the enzyme in human erythrocytes and homogenates of rat brain, kidney, lungs, liver and muscle tissue. Kinetic constants determined by this system for erythrocyte eholinesterase are presented. The data obtained with acetylthiocholine as substrate are similar to those with acetylcholine.

Synthesis and biological (in vitro and in silico) screening of the 4-aryl-fused pyranopyrazole derivatives as enzyme (α-amylase, α-glucosidase, acetylcholinesterase & proteinase) inhibitors with anti-oxidant and cytotoxic activities
2024, Journal of Molecular StructureThe development of single compound targeting dual/multiple proteins is an efficient approach to avoid the combined drug administration. Herein, based on the modification of the previously reported lead compound A, we synthesized a new series of dihydropyranopyrazolone 7a-g and dihydro-pyrazolopyranopyrimidin-5-amine 8. The compounds were evaluated for their inhibitory in vitro enzymatic potential against α-amylase, α-glucosidase, acetylcholinesterase & proteinase. Also, the in vitro antioxidant evaluation and cytotoxicity assays were performed for all the final compounds. Interestingly, compound 8 possessed moderate activity against α-amylase (25.38 %) and α-glucosidase (29.37 %) compared to acarbose (75.43 % and 52.79 %, respectively). On the other hand, compound 7f showed a moderate anti-cholinesterase activity (46 ± 0.03 % inhibition) compared to donepezil used as a reference standard drug (67.28 ± 0.61 %). Compound 7d exhibited a moderate anti-arthritic activity (proteinase denaturation (42.21 ± 0.04 %), inhibition of proteinase (38.09 ± 0.04 %)) compared to diclofenac sodium used as a reference (47.93 ± 0.01, 44.96 ± 0.02, respectively). Compound 8 exhibited significant antioxidant activity in TAC assay and increased ABTS scavenging activity. Compounds 7 and derivative 8 exhibited reduced or no toxicity against the normal skin fibroblast thus confirming their safety at least in vitro. The in silico ADMET (absorption, distribution, metabolism, elimination and toxicity) profile of all final compounds implied their acceptability as oral bioavailable drug-likeness property. The anti-hyperglycemic activity was emphasized by the molecular docking of compound 8 showing good interactions in the acarbose binding site of both α-amylase and α-glucosidase enzymes. This study presents the dihydropyrazolopyranopyrimidin-5-amine derivative (compound 8) as a promising scaffold for future modification to design and develop derivatives with increased inhibitory effect against α-amylase and α-glucosidase for the management of type 2 diabetes mellitus (T2DM).
Development of promising acetylcholinesterase and butyrylcholinesterase inhibitors: Synthesis, in vitro and in silico approaches of pyridine derived fused bis-oxadiazole and bis-thiadiazole derivatives
2024, Journal of Molecular StructureThe present study aims to synthesize new thiadiazole/oxadiazole fused thiadiazole derivatives, incorporating pyridine moiety as effective inhibitors of acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE) to treat Alzheimer disease. Initially, pyridine based thiosemicarbazone was synthesized by treating pyridine 2,6-dicarboxaldehyde with thiosemicarbazide. Thiosemicarbazone was cyclized into bis-thiadiazole, which further react with different isothiocyanates to give pyridine-based bis-(thiadiazole-fused-thiadiazole) derivatives (4a-h). Next pyridine based semicarbazone was synthesized by treating pyridine 2,6-dicarboxaldehyde with semicarbazide. Then semicarbazone was cyclized into bis-oxadiazole which further react with different isothiocyanates to give pyridine-based bis-(oxadiazole-fused-thiadiazole) derivatives (7a-h).The synthetic analogs were structurally elucidated through 1H NMR,13C NMR and HRMS (EI) analysis and assessed for acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE) inhibitory potential. All synthetic analogues showed varying degrees of inhibitory effects on AChE and BuChE, with IC50 value ranging from 12.89 ± 1.10 µM - 61.76 ± 2.40 µM for AChE and 14.79 ± 1.20 µM - 56.87 ± 2.50 µM for BuChE in comparison of standard inhibitor Galantamine (IC50= 37.40 ± 1.20 µM for AChE and IC50 = 41.20 ± 1.90 µM for BuChE). Among the series, analogues 4c IC50 = 13.32 ± 1.30 μM (against AChE), and 15.17 ± 1.40 μM (against BuChE), and 7c IC50 = 12.89 ± 1.10 μM (against AChE) and 14.79 ± 1.20 μM (against BuChE) exhibited notable inhibitory activity against AChE and BuChE enzymes, Moreover, molecular docking studies were conducted on potential analogues to investigate their binding interactions with the targeted enzyme's active sites.
New benzimidazole based Schiff bases as potent anti-alzheimer agents: Synthesis, bio-evaluation and molecular docking study
2024, Journal of Molecular StructureIn search of potent anti-Alzheimer agent benzimidazole based Schiff base derivatives (1–18) were synthesized and evaluated as dual inhibitor for acetylcholinesterase and butyrylcholinesterase enzymes. All analogs among the series except analog 1 and 16 showed a variable degree of inhibitory activity with IC50 value ranging between 0.10 ± 0.01 to 12.40 ± 0.30 µM for acetylcholinesterase and 0.20 ± 0.01 to 11.10 ± 0.30 µM for butyrylcholinesterase. The most potent analog found among the series was analog 8 having IC50 value 0.10 ± 0.01 and 0.20 ± 0.01 µM for both acetylcholinesterase and butyrylcholinesterase inhibition respectively. The structures of all synthesized analogs were confirmed through NMR and HR-EIMS. Structure activity relationship (SAR) has been established for all newly synthesized derivatives. To understand the binding interaction of most active derivatives with enzyme active site, molecular docking study were performed. The toxicity and mutagenicity of compound 8 was predicted using in silico software, namely Derek Nexus® (version 6.3). Various toxicity endpoints, including chromosomal damage, skin sensitization, hepatotoxicity were predicted. The degradation profile of compound 8 was predicted in silico by Zeneth software (version 9.0.1) resulting in a probability of the formation of seven potential degradation products.
Indolyl imine compounds as multi-target agents; synthesis, antidiabetic, anticholinesterase, antioxidant activities and molecular modeling
2024, Journal of Molecular StructureA new range of indolyl imine system 3d-l has been successfully prepared from 4,6-dimethoxy-2,3-diphenyl-indole-7-carbaldehyde 2a and 4,6-dimethoxy-3-aryl-indole-7-carbaldehyde 2b-c via Schiff base reaction. The structure of targeted compounds was confirmed by 1H and 13C NMR, FT-IR, mass spectrometry and single crystal X-ray diffraction techniques. The indolyl imine derivatives were also subjected to in vitro antidiabetic activities employing α-glucosidase and α-amylase enzymes. In terms of antidiabetic investigation, the α-glucosidase enzyme was found to be potential target due to the comparable inhibition concentrations with the standard acarbose and the compound 3e exhibited better potency than the standard. The anticholinesterase potency of the compounds was investigated towards the acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) enzymes. The compounds displayed moderate efficiency against the BChE enzyme with the best inhibition concentration of 30.48 μM by the compound 3h. The antioxidant properties of final compounds were determined by DPPH radical scavenging, ABTS Cation Radical Decolarization and CUPRAC Cupric Reducing Antioxidant Capacity assay methods. The ABTS cation scavenging assay provided the best responses for the compounds and the candidates 3k and 3l were determined as promising targets for the antioxidant activity. Plausible binding mode and interaction of ligands with the selected enzyme have been studied by molecular docking, supporting the experimental results. In silico ADME showed high drug likeness of the synthesized compounds.
Evaluation of antioxidant, antimicrobial, enzyme inhibition activity, and cell viability capacity of Hypericum heterophyllum vent., an endemic species in Turkey's Flora
2024, Journal of Molecular StructureThe aim of this study was determined the antioxidant, antimicrobial, enzyme inhibition activity, and cell viability capacity of H. heterophyllum (endemic species in the Flora of Turkey) extracts. H. heterophyllum methanolic extract was used in this study. H. heterophyllum plants collected at the beginning of flowering and during full flowering. P content was determined with calorimetrically in spectrophotometer. K, CA, Mg, Fe, Mn, Zn and Cu was determined with Atomic Absorption and N concentration were determined according to the Kjeldahl distillation method. H. heterophyllum (endemic species in the Flora of Turkey) extract was evaluated for its antioxidant (DPPH, ABTS, total phenol and flavonoid content), antimicrobial (four bacteria (Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, Bacillus cereus) and two Candida species (Candida albicans and Candida tropicalis)), enzyme inhibition activity (ɑ-glucosidase, ɑ-amylase, acetylcholinesterase, butyrylcholinesterase, tyrosinase), and cell viability (XTT assay) capacity. Extracts from the beginning of flowering period showed stronger biological activity. It has been determined that the extracts have antioxidant, antibacterial (against Staphylococcus aureus) anticancer activity (on breast cancer cells), and inhibition effects of BChE and alpha-glucosidase. It has been determined that the extracts obtained from H. heterophylum have important pharmacological activities, and the plant exhibits higher activity in the BF period. Hypericum heterophyllum Vent. The activities of the chemicals contained in it against various proteins that are α-gly enzyme (PDB ID: 1R47), AChE enzyme (PDB ID: 4M0E), BChE enzyme (PDB ID: 5NN0), alpha-Amylase (PDB ID: 3BAJ), breast cancer protein (PDB ID: 1A52 and 1JNX), were compared. ADME/T analysis of molecules with high activity was performed.
Monitoring acetylcholinesterase (AChE) is crucial in clinical diagnosis and drug screening. Traditional methods for detecting AChE usually require the addition of intermediates like acetylthiocholine, which complicates the detection process and introduces interference risks. Herein, we develop a direct colorimetric assay based on alkaline iron formate nanosheets (Fe(HCOO)2.6(OH)0.3·H2O NSs, Fef NSs) for the detection of AChE without any intermediates. The as-prepared Fef NSs exhibit oxidase-like activity, catalyzing the generation of O2·-, 1O2 and ·OH, which leads to a color change from colorless to blue when exposed to 3,3′,5,5′-tetramethylbenzidine. AChE directly inhibits the oxidase-like activity of Fef NSs, resulting in a hindered color reaction, enabling the detection of AChE. The biosensor has a linear detection range of 0.1–30 mU/mL, with a minimum detection limit of 0.0083 mU/mL (S/N = 3), representing a 100-fold improvement in detection sensitivity over the traditional Ellman's method. Satisfactory results were obtained when analyzing real AChE samples. Attractively, a method for the quantitative detection of AChE by a smartphone is established based on the Fef NSs. This method enables instant acquisition of AChE concentrations, achieving real-time visualized detection.
