Abstract

The vertebrate nervous system forms by the specification of, successively, neuroepithelial regions and cell groups. One of the proposed major histogenetic steps is the subdivision of the neural tube in compartments along its caudorostral and dorsoventral axis. This event is reflected, and may be directed, by the restricted expression of many transcription factors. Here, we report on the isolation of a new homeobox gene of thepairedsuperclass,Otlx2,whose early expression pattern in the mesencephalon and prosencephalon is congruent with proposed neuromeric models of brain morphogenesis. In addition, its late embryonic and postnatal expression, in clear continuity with the earlier pattern, suggests a role in the neuronal differentiation and the histogenesis of several prosencephalic and mesencephalic areas. Finally,Otlx2is expressed from the earliest morphogenetic events in the Rathke's pouch, the anlage of the adenohypophysis, an expression site shared by a very close homologue,Otlx1/Ptx1/P-Otx.

  • Transient Nodal Signaling in Left Precursors Coordinates Opposed Asymmetries Shaping the Heart Loop

Quantifications of the contribution of different factors to the fine shape of the heart loop, not just its direction, are expected to provide novel perspectives in understanding the origin of severe congenital heart defects associated with heterotaxy or other types of cardiac chamber misalignment. As Hesx1-cre also promotes recombination in the hypothalamus [17,18], SOX2 may be important for signals emanating from the ventral diencephalon that affect pituitary gland development; however, Bmp4, Fgf8, and Fgf10 were normal in this model. This bicoid homeodomain transcription factor is expressed in pituitary, eye, brain, tooth, heart, mandible, and umbilicus [19–23]. PITX2 (a.k.a. RIEG1, PTX2, OTLX2) is expressed in the oral ectoderm by e8.5 in mice, before the formation of Rathke’s pouch [19,24].

  • The lim homeodomain transcription factor LHX6 a transcriptional repressor that interacts with pituitary homeobox 2 (PITX2) to regulate odontogenesis

    2013, Journal of Biological Chemistry
    Citation Excerpt :

Identification of LHX6 expression in the oral and dental epithelium suggests that other factors, such as PITX2, might regulate Lhx6 expression. PITX2 is the first transcriptional marker of tooth development and is specifically expressed in the oral and dental epithelium (20, 21). A previous report on the role of PITX2 in craniofacial development using targeted deletion of Pitx2 isoforms in the mouse revealed decreased Lhx6 expression in the maxillary primordial of the Pitx2 null mutant mice (40).
