Restraint stress also robustly excited orexinergic neurons by improving calcium signals in a fiber photometry study (Gonzalez et al., 2016).

Central application of orexin A and, to a lower extent, orexin B leads to an increased secretion of corticotropin and increased anxiety-like behavior, which normalizes after administration of an orexin receptor 2 antagonist (Chang et al., 2007; James et al., 2017). Blunted orexin levels are predominantly assumed as a risk factor for mood disorders and suicidal behavior (Deats et al., 2014; James et al., 2017), but there are inconsistencies across the findings of different approaches.
