Consistent with earlier reports (Billups, 2005; Blaesse et al., 2005), we observed similar patterns of VGLUT1 and VGLUT2 immunoreactivities, indicative of the calyx-type giant presynaptic terminals surrounding the MNTB principal neuron cell bodies, which overlapped with the majority of Syp-positive presynaptic structures in wild-type preparations (Figure 1A, upper panels). Although merged images clearly showed substantial populations of vesicle clusters that were dominated by one of the two VGLUT isoforms, line-scan analyses of several different portions of the presynaptic terminals demonstrated that each vesicle cluster that was double positive for Syp and one of the two VGLUT isoforms contained detectable levels of the respective other VGLUT isoform (Figure S1A).

2017, Hearing ResearchCitation Excerpt :Projections into the VNTB are similarly heterogeneous, with terminals identified as glutamatergic (Billups, 2005; Blaesse et al., 2005), noradrenergic (Wang and Robertson, 1997), and serotonergic (Woods and Azeredo, 1999). Glutamate is the primary excitatory neurotransmitter utilized in the IC (Adams and Wenthold, 1979), and terminals in the VNTB are strongly labelled for VGlut2 but not VGlut1 (Billups, 2005). Using a combination of electron and confocal microscopy we were able to confirm that boutons from IC cells synapse directly onto MOC efferents in normal hearing mice.
