Experimental procedures
Procedures involving animals were in compliance with the National Institutes of Health Guide for the Care and Use of Laboratory Animals and were approved by the Institutional Animal Care and Use Committee. All efforts were made to minimize animal suffering, and only the number of animals was used that was necessary to generate reliable data. Expression pattern of BC1 RNA in the adult rat nervous system
We analyzed the distribution of BC1 RNA in the adult rat brain by in situ hybridization, using radiolabeled probes specific for the 3′ unique region of the RNA. The results are presented in the form of film autoradiographs for overview and in the form of emulsion autoradiographs for detail. BC1 RNA has been implicated in the regulation of local protein synthesis in postsynaptic microdomains in neurons,3 a mechanism now recognized to play an important role in the implementation of long-term synaptic plasticity.12., 29., 32. However, such mechanisms are not likely to be implemented uniformly in all neuronal subtypes as various types of neurons can be expected to differ from each other in their capacity to modulate postsynaptic form and function. It has long been known that these RNAs are abundant in well-defined regions of the brain (such as the hippocampus and cortical neurons) and only poorly detectable in other areas of the central nervous system [48]. The very specific subcellular localization of these RNAs (BC200 and BC1 are delivered to synaptodendritic compartments of neurons) together with their reported interaction with the Fragile X mental retardation protein, FMRP (a translational inhibitor of mRNAs localized at synapses [49], whose loss leads to mental retardation), led investigators to postulate a role for BC1/BC200 RNAs in the regulation of protein translation in the brain.
