Severe tactual memory deficits in monkeys after combined removal of the amygdala and hippocampus

Abstract

Monkeys with bilateral removals of both the amygdaloid complex and hippocampal formation were far more severely impaired on a tactual memory task than were monkeys with removal of either structure alone. These data parallel earlier findings on visual memory in monkeys and suggest that: (i) the memory deficit following combined ablations of the amygdala and hippocampus is multimodal; and (ii) the global anterograde amnesia observed in patients with medial temporal-lobe damage is also due to combined damage to these two structures.

      The nonhuman primate has been a productive model for the study of memory, establishing that long-term visual recognition relies upon cortical areas in the medial temporal lobe (MTL), particularly the rhinal cortex (Mishkin, 1982). This finding was soon extended to tactile memory (Murray and Mishkin, 1983), setting the expectation that the MTL may also support auditory LTM. To test whether selective ablation of the MTL disrupted auditory LTM, monkeys were trained to perform an auditory variant of the classic test for recognition memory, delayed-match-to-sample (DMS).

      Studies of auditory memory in nonhuman primates consistently report extremely slow learning of the rule for delayed match-to-sample (D’Amato and Colombo, 1985; Fritz et al., 2005; Wright, 1999) and short sample-stimulus forgetting thresholds (∼30 s), whether the task utilizes only two sounds (Colombo et al., 1996) or trial-unique sounds (Fritz et al., 2005). These findings suggest that although monkeys are easily able to form long-term memories in vision and touch (Mishkin, 1978; Murray and Mishkin, 1983), they may be unable to do so in audition, and are therefore limited acoustically to short-term memory (Fritz et al., 2005). More recently, we obtained evidence that even this type of auditory memory in the monkey is sharply limited (Scott et al., 2012), as it is likely to be dependent on a passive form of short-term memory (pSTM).
