2022, Experimental Cell ResearchCitation Excerpt :Proteins were extracted from GSCs or parental glioma cells using urea lysis buffer (8 M urea, 2 M thiourea, 4% CHAPS in 150 mM Tris-HCl) and separated by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) as described previously [16]. Electro-transferring of separated proteins onto polyvinylidene fluoride (PVDF) membrane was performed in a cool wet tank using Bolt and Mahoney buffer [17]. After blockade of non-specific reactivity by incubating with 3% BSA or 5% skim milk at room temperature for 1 h, the membrane was probed with the desired concentration of primary antibodies at 4 °C overnight.
